The recombinant genes were expressed in the Escherichia coli expression host, BL21(DE3), harvested as inclusion bodies, extracted into a urea buffer and purified. The radioactivity was measured on a gamma counter (Packard Cobra 5010) and Teicoplanin diluted to 25,000 cpm/μL in PBS containing 2% ethanol and 0.1% azide, and stored at 4°C. The measurement of pMHC-I stability was done as recently described [[14]]. Briefly, recombinant, biotinylated MHC-I heavy chain molecules in 8 M urea were diluted 100-fold into PBS buffer containing radiolabeled β2m and peptide to initiate pMHC-I complex formation.
